I have recently been experimenting with JQuery and one thing I have to say I'm impressed with is the ability to simply and concisely select certain DOM elements through a series of chaining methods and filters. Then, changing the HTML markup is as simple as appending / prepending more DOM elements. If you are familiar with JQuery at all is Dojo similar in structure to how it handles DOM element selection/manipulation? Would you recommend your book as a primary resource for this basic task?

Yes, Dojo has a similar query engine called dojo.query, which works in a similar vein. I don't cover it in my book, because I don't work in that way,using custom widgets with their own templates.
here's a good reference (with comparison to jQuery and others);
