We are in the process of implementing this forums software to A LOT of users (400,000+) and want to know if its possible to assign users to certain groups and other to other groups.

There are a bunch of ways to approach this but your description of what you need is kind of sketchy... can you elaborate on this? E.g.:

This is a one time, populate all the users and groups thing....

Or we've got an authentication system we want to integrate so changes to it's groups are reflected...

Or we want to identify users as they register or come in via SSO....
