On the Android site in Icon Guidelines it states:
Icons should make use of the alpha channel, and should not simply be full-frame images. Where appropriate, distinguish your icon with subtle yet appealing visual treatment.
How do I achieve this in Paint

If you mean MS Paint, then you can't. You will need to us an application that understands transparency, which Paint does not. Something like Gimp (free).
